Culture medium	Improved GAM medium (mGAM): peptone 5.0g,䏡 Peptone 5.0g, soybean peptone 3.0g, yeast powder 2.5g, beef powder 2.2g, digestive serum powder 10.0g, beef liver powder 1.2g, glucose 0.5g, potassium dihydrogen phosphate 2.5g, sodium chloride 3.0g, soluble starch 5.0g,L-cysteine 0.3g,L-arginine 1.0g,L-tryptophan 0.2g, sodium thioglycolate 0.3g,PH 7.3±0.1,121 ℃,15min. note: (add 1ml of sterile 0.1% vitamin K1 solution and 2mg of sterile hemin for every 200ml, and mix well). preparation of sterile hemin solution: weigh appropriate amount of hemin, add appropriate amount of distilled water, and sterilize under high pressure at 116 ℃ for 20-30 minutes.
Subculture procedure	(1) Prepare a test tube containing 5~10mL of liquid medium and 2 plates; (2) Open it in the biosafety cabinet, heat the tip of ampoule in a flame, quickly drop sterile water to creak it, then break it with forceps; (3) draw 0.5mL of liquid culture medium into a freeze dried ampoule, fully rehydrate and transfer the solution to the liquid test tube, mix evenly; (4) inoculate a plate with 0.2mL of suspension liquid, repeat the step to obtain two plates; ⑤ Put all the liquid test tubes and plates under the above culture conditions for cultivation, and the strains can be used when they grow.

Handling instructions of agar plate is enclosed. Please operate in strict accordance with this instruction, otherwise the replacement of bacteria are not be available in case of aberrant growth and loss of viability.

Growth conditions：37 ℃, anaerobic, modified GAM agar, 4-5 days, modified GAM agar: monthly peptone 15.0g, tryptone 10.0g, soybean peptone 3.0g, yeast powder 5.0g, beef powder 2.0g, beef liver powder 1.2g, soluble starch 0.3g,L-cysteine 0.15g, glucose 3.0g, potassium dihydrogen phosphate 2.5g, digested serum powder 13.5g, sodium thioethanolate 0.15g, sodium chloride 3.0g, agar 13.0g (not included in liquid medium), distilled water 1L,pH 7.0 ±0.2, 25 ℃ additive: 0.02% vitamin K1 solution and hemin

Recovery steps:

(1) Prepare a test tube containing 5-10mL of liquid culture medium and 2 plates;

(2) Open the ampoule in the biosafety cabinet, heat the tip of ampoule in a flame, quickly drop sterile water to creak it, then break it with forceps;

(3) Draw 0.5mL of liquid culture medium into a freeze dried ampoule, fully rehydrate and transfer the solution to the liquid test tube, mix evenly;

(4) Inoculate a plate with 0.2mL of suspension liquid, repeat the step to obtain two plates;

(5) Put all the liquid test tubes and plates under the above culture conditions for cultivation, and the strains can be used when they grow.

Recovery record: According to the recovery instructions, the results of the recovery are reported as follows:
