Culture medium Nutritional gravy medium (NA/NB): beef paste 3.0g, peptone 10.0g,NaCl 5.0g, agar 20.0g (without liquid medium), distilled water 1.0L,pH 7.0. Sterilization at 121 ℃ for 15min. [Note] Adding 5 mg MnSO4 · H2O to culture Bacillus is beneficial to produce spores.

Growth characteristics Gram-negative bacilli, with a size of 0.6-2.0 μm × 1.0-4.0 μm, usually move with perennial flagella; facultative anaerobic, low nutritional requirements; citrate cannot be used as the only carbon source; gas production, most strains do not produce H2S, ferment arabinose instead of xylose, which is only pathogenic to humans and can cause symptoms similar to typhoid fever.

Receiving notice: if any abnormality is found on the day of receipt, please contact the customer service within 24 hours. The bacterial tablets shall be used up once and shall not be retained once the penicillin bottled is opened. Keep it at -20℃ for storage if it is not used.  Please operate in strict accordance with this instruction, otherwise the replacement of bacteria are not be available in case of aberrant growth and lose of viability.

Medium : nutrition gravy medium beef paste 3.0g, peptone 10.0g,NaCl 5.0g, agar 20.0g (without liquid medium), distilled water 1.0L,pH7.0. Sterilization at 121 ℃ for 15min.

Recovery steps:

(1)Prepare a sterile test tube of 5~10ml liquid medium and 2 pieces of 90mm agar plates;  
(2)Sterilizing of the ampoule, under aseptic conditions, remove the cap and take out the inner vial. lift the cotton plug, rehydrate the pellets by drawing 0.5ml of liquid culture medium into the vial.
(3)Fully dissolved, transfer the solution to the liquid test tube, mix evenly; 
(4)Inoculate a plate with the solution, distribute it well in 200ul/ plate; the plates are not allowed to seal by wrapping. 
(5)Put all the plates and test tubes under the above culture conditions for cultivation, The culture solution is obviously turbid after 18-24 hours, and this indicates the bacterial grow well.
(6)If the strain does not grow, it should be continued to be cultured for 72h, and then used according to the test requirements.
