2. Storage conditions
Storage of freezed dried ampoule and agar slant at 2°C to 8°C
3. Growth Conditions
1, 112 Van Niel ' S Yeast Agar:K2HPO4 1.0 g,MgSO4 0.5g, yeast extract 10.0 g, distilled water 1000 ml,pH7.0-7.2. 121 ℃,15min.
2.Atmosphere: anaerobic, cultured under tungsten lamp
3. Temperature: 30 ℃
4.Culture method: fill 15-20mL screw test tube with liquid culture medium and sterilize for later use. Inoculation: In the safety cabinet, after absorbing 0.3-0.5mL of culture medium to dissolve the bacterial powder, all of them are beaten back into the liquid culture medium and sealed tightly. Let it stand in the incubator, illuminate the incubator for 5 days, observe the turbidity of the liquid, and the color is dark red, that is, the activation is successful.
4、Notes:
1.Normal culturing time, 1-2 days for bacterial, 3 days for yeast, 5-7days for mould, 7-10days for fungal.
2.Agar slant shall be inoculated asap, and do not keep the storage for more than 3 months.
3.Please recover the strains in strict accordance with this instruction, otherwise the replacement of the strain are not be available in case of viability loss caused by different media or growth conditions.4.Waste generated from the handling process should be discarded after high-pressure sterilization.
