Culture medium Malt juice agar (MA): malt extract 20.0g, glucose 20.0g, peptone 1.0g, agar 20.0g, distilled water 1.0L,pH natural. Sterilization at 121 ℃ for 15min.
Subculture procedure (1) Prepare a test tube containing 5~10mL of liquid medium and 2 plates; (2) Open it in the safety cabinet, heat the tip of ampoule in a flame, quickly drop sterile water to creak it, then break it with forceps; (3) draw 0.5mL of liquid culture medium into a freeze dried ampoule, fully rehydrate and transfer the solution to the liquid test tube, mix evenly; (4) inoculate a plate with 0.2mL of suspension liquid, repeat the step to obtain two plates; ⑤ Put all the liquid test tubes and plates under the above culture conditions for cultivation, and the strains can be used when they grow.

Growth characteristics The colony texture is fleece; the color of the colony is yellowish brown, and the old back edge becomes dark golden yellow; the reverse side is purple-red; the colony is flat, the entire edge, and the center point is convex; the exudate is a small amount of transparent small droplets at the beginning, and then becomes Larger lavender-red droplets. Growth conditions:28-30 ℃, aerobic, integrated PDA. Comprehensive PDA: potato boiling juice 1000mL, potassium dihydrogen phosphate 3.0g, magnesium sulfate heptahydrate 1.5g, glucose 20.0g, vitamin B1 10mg, agar 20g,pH natural. 121 ℃,15min. Potato boiling solution: 200g of potato, peeled, cut into pieces, boiled in distilled water for 30min, constant volume of filtrate to 1000mL for later use. 121 ℃,15min.

Recovery steps:

(1)Prepare 1-2 pieces of SDA plates; 

(2) Open it in biosafety cabinet, heat the tip of ampoule in a flame, quickly drop sterile water to creak it, then break it with forceps; 

(3) Open the ampoule in the biosafety cabinet, heat the tip of ampoule in a flame, directly dispense 200ul of the liquid suspension into a SDA agar plate evenly.

(4) Put the plates under the above culture conditions for cultivation for 5-7 days.
