Culture medium Nutritional agar chocolate plate (English name: NA-C): beef paste 3.0g, peptone 10.0g,NaCl 5.0g, agar 20.0g, distilled water 1.0L,pH 7.0. After sterilization at 121 ℃ for 15min, cool to 80-90 ℃, add 5% sterile defibrated sheep blood, shake well and pour the plate to cool for later use.
Subculture procedure Take freeze-dried powder and coat two plates on chocolate plates for the first time, culture for 48-72 hours, the plates are covered with bacteria layer, then transfer and coat five chocolate plates, each plate is eluted with 3mL PBS after culture for 72 hours, the eluted bacteria solution is about 10mL for later use, the concentration of bacteria suspension is about 109CFU/mL, diluted to 10-6, 10-7 and 10-8 gradients, and 200 μL of coating is taken for counting, select plates with colony numbers between 30 CFU and 300 CFU for counting;
